A study designed to determine the association between the level of human milk oligosaccharides at six weeks after birth and anthropometric characteristics observed in human milk-fed infants up to four years old.
In a longitudinal cohort study of mothers, 292 samples of their milk were collected. The mothers were, on average, 60 weeks postpartum, with a range from 33 to 111 weeks. Among the infants, a group of 171 were exclusively breastfed until the age of three months, and an additional 127 maintained this exclusive breastfeeding regimen up to six months. High-performance liquid chromatography served to quantify the concentrations of 19 different HMOs. By quantifying 2'-fucosyllactose (2'FL), the maternal secretor status (n=221 secretors) was determined. Z-scores for weight, length, head circumference, the sum of triceps and subscapular skinfold measurements, and weight-for-length were calculated across the 6-week, 6-month, 12-month, and 4-year time points. Employing linear mixed-effects models, we analyzed the correlation of secretor status with each HMO metric and how they changed from birth for each z-score.
Anthropometric z-scores, up to four years, remained unaffected by the maternal secretor status. At both 6 weeks and 6 months, specific HMOs displayed z-scores, noticeably within subgroups characterized by secretor status. Children born to secretor mothers exhibiting higher 2'FL levels demonstrated increased weight (0.091 increase in z-score per SD increase in log-2'FL, 95% CI (0.017, 0.165)) and length (0.122, (0.025, 0.220)), but no corresponding changes in body composition measures. Weight and length gains were significantly more pronounced in children of non-secretor mothers, displaying a positive association with increased levels of lacto-N-tetraose, according to the statistical analyses. We explore the long-term consequences of maternal bisphenol A (BPA) exposure during pregnancy and breastfeeding on ovarian function in adult mothers as they transition to older age. The developmental progression of follicles within BPA-exposed ovaries was impeded, with growing follicles arrested at preliminary stages, thus hindering their maturation to the mature stage. Follicles undergoing atresia, and those in the early stages of atresia, also experienced enhancement. An impairment in estrogen and androgen receptor signaling was detected within the follicle population of BPA-exposed females, characterized by a high expression of ER and a greater prevalence of early atresia in mature follicles. Ovaries exposed to BPA displayed a greater presence of the ER1 wild-type isoform, distinct from its variant isoforms. Exposure to BPA influenced steroidogenesis by reducing the production of aromatase and 17,HSD, and conversely increasing the production of 5-alpha reductase. The serum levels of estradiol and testosterone decreased in BPA-exposed females, mirroring this modulation.
